The 19th century values of work and of being an independent business man clashed with the modern 20th century values of extreme expansion with large work forces and of earning the most money possible. The rise of the robber barons and the captains of industry helped the economy by pushing America into first place in the production of several products and by creating many new jobs. Although these new opportunities appealed to the masses, not everyone was satisfied by his new occupation. The creation of labor unions was a reaction to the numerous complaints about working conditions, wages, and work hours. The first unions protested with peace and reason. Once they realized that nothing could be accomplished through negotiation, drastic measures were taken and violence was the answer to their problems. The clashes between management and workforce in the Great Railroad Strike, Homestead Strike, and Pullman Strike emphasize these crises that were resolved through force and destruction. Economic depression in 1873 was the main factor in setting off the 10% wage cuts and shortening of work days in the railroad business. In 1877, Laborers took action by seizing control of the rails by sequestering the rail switches and by blockading freight trains, only letting passenger trains through. Strikes broke out in many cities including Baltimore where 10 protesters and bystanders were killed by the local militia. Engulfed in rage, the laborers rampaged through the city destroying all things pertaining to trains. Only after Hayes was called for help did the real action begin. In Pittsburgh, the National Guard was called to quell another strike by any means necessary, even if it meant killing 20 people including women and children. Tempers exploded as fires raged through the city destroying $4 million worth of railroad property. The country suffered an estimated 100 deaths due to these rash actions and violence. The casualties could’ve been avoided through peaceful negotiation, but that never worked. Carnegie’s Homestead Steel mills, a part of US Steel, were the center of a hostile conflict in 1892 between laborers and management caused by large wage cuts and short work weeks. These cuts were commanded by Frick who reasoned that increased efficiency inflated salaries. Frick closed the mill and hired and placed 300 Pinkerton Detectives on guard duty. On July 6th, much blood was spilled as a gunfight between the guards and laborers ensued for 12 hours. The workers won the battle, but lost the war completely. ,500 National Guard members took control of the mill and the blame of an assassination attempt at Frick was blamed on the Labor Unions. In November, all steel unions were broken, and the mill operated on cheap labor from African Americans and immigrants. The hours were brutal as they were pushed to work 12 hours a day with a 24 hour day every 2 weeks. The Steel Industry didn’t see unions for another 44 years. The management had won. The government was an avid supp orter of big business which is evident in the Pullman Strike of 1894. Still feeling the effects of depression, the Pullman Palace Car Company had to lay off 5,800 workers and also cut wages from 25-50%. Even though they were making less money, the Pullman town didn’t lower the rent. On May 11th, 90% of the workers went on strike and a month later, ? million across the country joined them by refusing to pull Pullman sleeping cars. Lawyers, as tricky as they are, managed to get a court injunction to force the workers back to work by attaching a US mail car to a Pullman car and say the laborers were disturbing the distribution of mail. The workers didn’t comply to with the courts jurisdiction and so 14,000 troops were sent by Grover Cleveland to enforce the ruling. 25 were killed as the troops shot into a crowd of 10,000 in Chicago. Hundred were jailed including Eugene Debs, leader of the American Railway Union who ordered the workers to stop pulling the Pullman cars. The government prioritized a strong healthy economy over a large and happy population. Rapid industrialization and nation-wide corporations led to wealthier families and a new middle class. Although there were many who did enjoy the new luxuries paid for by new salaried jobs like managers, technicians, and engineers, many more people were below the poverty line and resided in the slums, living with other families with no privacy and unsanitary conditions. These families were unfortunate because of the wage cuts and the replacement of labor by immigrants and African Americans. A change was in order and the labor unions took the plate by voicing their complaints, and, when that didn’t work, resorting to physical means to get what they wanted.

Increasingly, judges are being encouraged to find consideration in cases; this is so that contractual claims can be assessed on the basis of the real intention of the contracting parties to one and another, rather than a strict and narrow interpretation of case law. Consideration is only at issue in simple contracts, Courts often have to find consideration to be able to infer that a contract exists. In a Contract By Deed, consideration is unnecessary; as it is clear what is due form whom. In addition, where the traditional narrow view of consideration may lead to inconsiderate results, Courts may accept a request for applying the principle of equitable estoppel. Therefore, a contract which contains promises unsupported by consideration is void ab initio. Valid consideration has the following features: 1. Consideration must move from' the offeree to the offeror, that is, the person making the offer must be expecting something in return. 2. Consideration must be something of value, however trifling to the offeror, or something of detriment to the offeree. 3. Consideration must be sufficient in law, but need not be sensible in fact. An offer of sale of a Rolls Royce Car for 1 is valid consideration though not sensible. However, if this car is offered gratis, there is no consideration and the agreement is unenforceable. 4. Proteins are the chain of amino acids which binds with polypeptide backbone and then fold in a unique 3D (native) structure by which protein expression takes place. Various forces and factors are responsible for protein folding .If right expression of the protein will not take place, it will cause disorders in human body. Many diseases like Alzheimers; Parkinson, cystic fibrosis etc (Baldwin 2007) is caused due to improper folding of proteins. For proper expression of protein, the amino acid chain should be in its unique 3D structure. Sometimes proteins require assistance in folding, molecules which help in the folding are known as chaperones. These molecules help in the folding of the certain protein molecules and also prevent the unfolding of the molecules. In this assignment, I have tried to describe the mechanism of the protein folding and effect of various factors which influence protein folding by taking into consideration the present developments in our understanding of thermodynamics and kinetics of protein. INTRODUCTION Protein folding refers to the process by which a protein assumes its characteristic structure, known as the native state. Protein folding is very complex mechanism and great development in its understanding has been achieved in last 20years due to the development and use of some sophisticated modern techniques like X Ray Crystallography, N.M.R, and Mass Spectrometry etc. In the last few decades, we have managed to find the 3D structure of various proteins and how actually protein folding takes place. In the complex process of protein folding, various factors act together to construct specific 3D structure of a protein. Protein folding is a very quick process taking milliseconds to seconds. Due to high speed of folding, it is not possible to find each and every possible conformation in fractions of time. Levinthal stated that each protein can possibly have millions of pathway by which a desired 3D pattern can be achieved. By study of intermediates we can understand the mechanism or pattern on which protein folding works. But it is impossible to find out all possible structure of protein molecules. Protein folding study uses denaturants which help to denature the protein. The logic is to use denaturing conditions viz. high pH, Temp, Pressure etc to stop folding at intermediate stages (Stop flow technique) to study these intermediates, thereby generating an overview of the whole process. Fully folded structure is also known as native structure. Protein folding studies have also been done in in-vivo and in-vitro conditions. In in- vivo protein folding, some metal ions act as cofactors. These cofactors stabilise and accelerate protein folding and finally help to achieve native stage .Macromolecules like chaperones initiate protein folding, helping the protein molecule to retain its folded confirmation and making it stable. In-vitro concentration of macromolecules is very less, around 1% of what is originally present in the cell. So inside cell, these molecules also affect protein stability and make them more stable than in vitro (Rumfeldt et al. 2008) Protein folding can be a two stages or multistage process, depending upon the type of sequence. A single point mutation can change folding from two stages to multistage and vice-versa .Some previous studies show that protein sequences of less than 80 amino acids prefer two stage folding and large protein sequences having more than 130 amino acids prefer multi stage folding. Later studies have shown that sequences rich in F and G amino acids prefer two stages folding where as sequences rich in C, H, L and R amino acids prefer multistage folding (Ma, Chen Zhang 2007) PROTEINS FOLDING Hydrophobic and hydrophilic interaction Proteins are made of amino acids and joined by polypeptide bonds leads to formation of polypeptide backbone. Hydrophilic and hydrophobic interactions of the amino acids adjust protein it such a way that it suffers minimum repulsion and problem from the surrounding. In folded state of protein all hydrophilic amino acid molecules come at the other side and interact with water in the cytoplasm and hydrophobic molecule come at the inner side of the molecule and dont show any reaction and attraction with water molecules and ultimately protein folding take place in such a way that suffer minimum repulsion (Baldwin 2007) All amino acids have different energetic in secondary structure. There are many chameleon sequences in protein structure which can be taken as alpha helix or beta sheets depending upon the tertiary structure of protein. These chameleon are stabilised by hydrophobic forces (Chen et al. 2008) Fig.a shows yellow colour hydrophobic molecules inner side of structure Fig.b shows folding takes place and keeps hydrophobic molecules in inner side (Chen et al. 2008) Apart from these interactions hydrogen bonding is very important in the protein folding. Hydrogen bonding takes place between hydrogen and electronegative atoms In the maintenance of the native structure hydrogen bonding present in between the polypeptide amino acid chain which helpful in the formation of the secondary, tertiary and quaternary structure of the proteins. Hydrogen bonding also interacts between polar and side chain residues with the surrounding water molecules. During the denaturing of the protein hydrogen bond between protein molecules break and native structure of protein disturbed (Djikaev, Ruckenstein 2010) Hydrophobic interactions are also responsible for the protein folding. During hydrophobic interactions amino acids which are non polar or hydrophobic they align themselves in such a way that all hydrophobic come together and all hydrophilic molecules make hydrogen bonds with water molecules, all hydrophobic amino acid come in to inner side of the protein molecules and formation of the nuclei take place which is hydrophobic these interaction further helpful in the secondary, tertiary structure and due to this protein sta bilise and help to achieve native structure because hydrophobic molecules will not interact with water and always have repulsive attitude toward water and let the protein in the folded form and finally such type of the folding take place in which molecule have hydrophobic core and all hydrophilic molecules in periphery of the folded structure(Berezovsky et al. 2001) Free energy and entropy (G, E) Gibbs free energy (G) is also defined as the amount of the energy which is free and this can also be defined as the measure of unstableness and this measured by simple equation G= H-TS G=free energy, H=enthalpy, S=entropy, T= temperature Entropy (S) is defined as degree of randomness in any system, in terms of protein folding this can be define as the measure of the possible structure in the protein molecule at that value of the entropy. Entropy is responsible for the possible out come of protein as much high would be entropy as much high number of the confirmation would form. To minimise the entropy in the protein folding various force come in play like hydrogen bonds, salt bridges, disulphide bonds etc. these bonds help to reduce the entropy and favour protein folding(Brady, Sharp 1997)BOLTZMANN worked on the entropy and give us a very useful relation between atomic theory and entropy. Boltzmann proposed an equation which shows that entropy in any unfolded protein structure is equal to the product of his constant(KB) and natural log of number of all possible states which any protein can adopt(S) S = KB * NATURAL LOG (S) KB=Boltzmann constant Protein folding prefers low value of entropy and follows that smallest path in which value of entropy is low. At different value of the entropy different number of structure could be present and finally all structure will vanished and one structure would be present at the minimum value of entropy, which is our native structure (Weikl, Dill 2003) Enthalpy (H) is also responsible for protein folding. This is also known as the measure of the total energy of the system including internal energy (U). P and V are the pressure and volume of system H= U+P*V So, we should search for such structure which have low value of enthalpy then the free energy, because which structure have low value of enthalpy that will have low value of free energy too(Brockwell, Smith Radford 2000) Gibbs free energy equation is the single solution for all protein folding problem. Chaperones molecules are task specific that are different molecules perform different functions. For example: HSP70/40 They prevent aggregation and misfolding of newly synthesised protein molecules. HSP60 They unfold intermediate and then fold them properly in to native structure Chaperones are like catalyst, they enhance rate of protein folding and assist protein folding to native structure and after the formation of the native structure they separated. As like catalyst they required energy for the initiation of the process Chaperones activity is much specialised, in stress condition protein get mutated, denature, and aggregate. Hsp90/70 mechanism chain Some chaperones may responsible for disease. it has been found that HSP90 enhance cancer development because many mutated protein mature in the presence of the HSP90 and cause cancer , repressing HSP90 such type of cancer can be able to prevent but problem associated with this is due to this action some non mutagenic protein will be degrade and not express and cause problem. But on other side HSP70 acts as good repressor in neurodegenerative disease and prevent this disease in the fruit flies (Tomala, Korona 2008) Models of protein modelling and structure prediction Plaxco and co-worker model: This model shows that high degree of correlation between folding rate and structural properties of protein explain on the basis of contact order (CO). They calculate ÃŽÂ ¦ value and on this basis they find high correlation between simulation and experimental ÃŽÂ ¦ value .in the end of their experimental model they conclude that L24 and G24 are two most important residues in the folding of proteins Physics and bioinformatics based models: Physics models are very helpful to understand protein folding rate and route to folding. These physics based models help to understand the various forces and their dynamics in protein folding. These models help to understand: Conformational changes in protein Mechanism of folding, enzyme catalysis, mode of action protein Response to ph salt and denaturants(Brockwell, Smith Radford 2000) Bioinformatics is very important tool to find out the structure and folding pattern of the protein molecules. In this we add our computer based program along with these physics model and within the fraction of time provide us 3D structure of protein. You will save yourself many future headaches. True, a few find their lifework accidentally, but there are hundreds who never find their right vocations at all. About 20 per cent of workers are occupational misfits. Jobs change rapidly. Employment opportunities are different from when Dad was a boy. They differ from year to year. In the world today, for example, there are more than 300,000 different occupatio ns.To find the right life- work you need to study your interests and abilities and to explore a variety of vocations. Although there are a vast number of occupations from which to choose, certain traits are required for success in any field-accuracy, ambition, loyalty, and perseverance, for example. Is your supply of perseverance low? Do you become discouraged if you don't succeed in something? What are you going to do about it? Select a vocation for which you are physically qualified. When young people enter a job or university/college, school officials are called upon to write letters of recommendation. What is your attendance record? Your school record? Do you participate in sports, other activities? Are you trustworthy? Even when there is a labor shortage, employers want to know these facts about their prospective employees. You are making now the record upon which the school's recommendation will be made.You are interested in a certain broad field of work it may be scientific, mechanical, or commercial. Reading can fill in the gaps in your job observation and experience. In every field there are jobs ranging from those requiring little training to those which require long training and constantly increasing skill. The jobs at the top are fewest. You may be an electrici an's helper or an electrical engineer; a draftsman or an architect; a filing clerk or a private secretary; a dressmaker or a dress designer. Nowadays, everyone has a smartphone, tablets, or any other kinds of mobile devices. And one of the most appealing traits of these devices is accelerating the dissemination of information and knowledge, which means you can grab any news you’re interested in anytime and anywhere. However, the mobile devices have also destroyed communication among our friends and family. For instance, when I went out with my friends and had a dinner with them, most of us just focused on the tiny little screens on our hands without talking to each other. Moreover, they chatted with each other via Facebook even when they just sat nearby. In other words, these mobile devices have not only improved the quality of communication, well†¦on the Internet, but also have destroyed communication among friends directly. Therefore, I think that we all need to figure out the original intentions that drives us want to use a smartphone, such as productivity improvement, joyfulness, or merely pursuit of vanity. No matter what the motivation it is, trying not to be obsessed with these gadgets is the most important thing to us. 3 THOUGHTS ON â€Å"SMARTPHONES AND TABLETS HAVE DESTROYED COMMUNICATION AMONG FRIENDS.† You’re right about this; it’s such a shame that people really don’t talk face-to-face anymore. I like to make a rule at family meals that we all put away the phones and gadgets for the time that we’re eating. It helps to make a shoebox or something that everyone can put their stuff in so everyone knows no one else is checking theirs. 😛 But, yeah, people have to learn again to ‘be with the people who are HERE,’ as the person who gave me the shoebox idea wrote on hers. Soren Kierkegaard Vs. Friedrich Nietzsche Free Online Research Papers Soren Kierkegaard and Friedrich Nietzsche are widely regarded as the two greatest thinkers of the 19th century. Known as the fathers of the existentialist movement, these two philosophers have revolutionized the way we see the world. Even though it has been over a century since they have both died, the very fact that their texts are being taught in modern curriculum’s, is a testament to how influential their works truly are. Though both men have similarities, in the sense that they wish to discover the true meaning of ones existence, they both come to two completely different conclusions. Though both state philosophy as a means to enrich and appreciate the life you possess, Religion, the major difference between these two thinkers, is what is wrong with their purposed ideas of living a meaningful life. Kierkegaard, in of his works Concluding Unscientific Postscript to Philosophical Fragments states: ‘Without risk there is no faith. Faith is precisely the contradiction between the infinite passion of the individual’s inwardness and the objective uncertainty. If I am capable of grasping God objectively, I do not believe, but precisely because I cannot do this I must believe. If I wish to preserve myself in faith I must constantly be intent upon holding fast to the objective uncertainty, so as to remain out upon the deep, over seventy thousand fathoms of water, still preserving my faith.’ One can wholeheartedly disagree with Kierkegaard, for the simple reason that faith is not a standard for belief. One does not need to believe, because they can not objectively deduce. Kierkegaard’s philosophical opinions were greatly shaped by his religious views, one of the most famous being his theory on The Knight of Faith. Kierkegaard defined the Knight of Faith as â€Å"the individual who is able to gracefully embrace life†. He is an individual who relies on acceptance of the absurd as a means of life. The Knight of Faith is willing to step over ethical boundaries for what he believes is a higher purpose. In his work Fear and Trembling Kierkegaard expands on this thought. Kierkegaard’s example of such a knight was the biblical figure of Abraham. When Abraham was asked to sacrifice his son Isaac, he wrestled with the decision. How could he take the life of his beloved son? His heart heavy with sorrow, Abraham took his child to the top of a mountain and raise d his hand to kill his son; at the last minute, the angel of the LORD prevented him from doing so. Instead of Isaac, Abraham sacrificed a ram that was trapped in a thicket nearby. Abraham was rewarded with the life of his son, because of his faith in the absurd. Kierkegaard claims Abraham, out of love was willing to kill, and because he was sacrificing his son to god, out of love, he is the true knight of faith. This is what is most confusing. Abraham travels to the top of the mountain to kill his son, believing it will all work out, based on nothing rational. Based on simple ethics, he is going to become a murderer. His loyalty and faith in god puts him above human ethics and morals. This notion is absolutely insane. This notion, that if an act is done out of love, and you receive a message from god, than your actions are suddenly justifiable. Kierkegaard believed Abraham such a hero, that he not only idolized him, but wept, because he did not believe his faith to be as strong as his. This perfectly connects to his theory of angst. This concept of angst (anxiety) is used to define the feelings of fear and insecurity of the free thinking human being. Kierkegaard meant this in regard to fear of failing in your responsibilities to god, although a person of none, or a different faith, can attribute the same principles to their fear of not living up to their own morals and beliefs. Simply replace God, for anybody whose opinion matters to you, your family, and friends, significant others. The problem of anxiety applies to these situations. You wish so hard to please these people, but the fear of disappointing them is always present in your mind. In his works Kierkegaard frequently speaks of the sub specie aeterni (from the perspective of eternity). It was in his opinion that ones life should be viewed from the perspective of eternity. Kierkegaard believed all humans were a synthesis of the finite (body) and the infinite (soul) Kierkegaard defined humanity as a tension between the finite and infinite, a tension that should not be disregarded but intensified. For example a guitar is not useful when silent; it’s at its most beautiful when being played, the strings vibrating. For Kierkegaard life was not meant to be lived in just a state of religious solitude, like a monk, one must suffer to experience true â€Å"tension†. Those who center their lives on being practical are missing out on spirituality. According to Kierkegaard these people feel hopelessness because they are not experiencing the spiritual aspect of the human experience, thereby unable to be fully human. Where Kierkegaard loses some of his readers is the notion that the finite and infinite synthesis can only occur through god. He lives by the idea that through god people can avoid depression, and this feeling of nihilism. In his text The Sickness unto Death Kierkegaard claims we are all in a state of despair. He states that society as a whole is failing to live up to the true human experience, even going so far as to state it is in fact a SIN in this condition once we have been taught the word of Christ. What if we aren’t in a state of despair? What if we don’t feel our lives are meaningless, and sinful? If someone was not of the Christian faith, perhaps Kierkegaard’s views on sin and despair were irrelevant in his/hers way of life. Without using faith or atheism to analyze his texts, one can gather from Kierkegaard’s works a simple message. Rely on no person or facts of this world to provide you with answers to philosophical and ethical questions. We are the ones who will have to live with the choic es we make. We will be held responsible by our consciousness four our ethical decisions. Therefore we should act according to our own personal beliefs: we should do what makes sense to us. Do we really need go to provide us our moral compass? The next philosopher being discussed would argue no. Friedrich Nietzsche was an atheist. This belief was a major influence in most of his works, going so far as to state â€Å"god is dead†. In his text Thus Spoke Zarathustra he states: God is dead. God remains dead. And we have killed him. How shall we comfort ourselves, the murderers of all murderers? What was holiest and mightiest of all that the world has yet owned has bled to death under our knives: who will wipe this blood off us? What water is there for us to clean ourselves? What festivals of atonement, what sacred games shall we have to invent? Is not the greatness of this deed too great for us? Must we ourselves not become gods simply to appear worthy of it? Nietzsche does not mean the literal death of god, but a metaphorical death. Nietzsche hoped that with the death of god, society would finally be able to create their own sense of morals and ethics. He hoped for people to lose their faith in god and come to the realization of nihilism. Defined as the philosophical doctrine suggesting the negation of one or more meaningful aspects of life, most commonly, nihilism is presented in the form of existential nihilism which argues that life is without objective meaning, purpose, or value. Nietzsche hope through nihilism humanity would be forced to re-evaluate their very foundations, foundations deeper than Christianity and other abrhamic religions. Understand that Nietzsche was not a fan of nihilism; in fact he was quite fearful of it. â€Å"I praise, I do not reproach, [nihilisms] arrival. I believe it is one of the greatest crises, a moment of the deepest self-reflection of humanity. Whether man recovers from it, whether he becomes master of this crisis, is a question of his strength! It is possible. . . . (Complete Works Vol. 13) He saw it as a stepping stone to reach the greater realization of what he believed to be the ultimate way of life, the Will to Power, to become the ÃÅ"bermensch. Back when the gangsters and racketeers ruled the music business, the music had a real sense of integrity to it. That was back when gangsters knew of "this hot new band/artist" they had noticed in their buddy's club. Then, if they owned their own club, they book that band for gigs. Or maybe a relative of theirs was in a band, and that band had street credibility; all they needed was a break. The bands took pride in making a full album, a collection of songs reflecting their current state of affairs. Back then the people who ran the record companies were music fans. In the last ten years, however, a disturbing trend has taken over. While all of the rockers were out entertaining the people, all of the scholars were off getting their business degrees. This led to a massive take-over. When the scholars finished their schooling, some of them used their business degrees to get into the music business. They became record label executives, and as a result, they viewed the record label as more of a bank then an entertainment industry. Of course, every label gets in the game to make money, but the simple fact is that music fans will buy whatever they are told to buy, for the most part. Gone are the days of bands having to prove themselves on the club scene, building their live performance and song repertoire. The music business is now a fast-food industry - 15 minutes of flash, here today gone tomorrow artists. The ones who suffer are the kids, but the target demographic that the labels go after are none the wiser. An at-home, online, record store that allows the fans to pick and choose their music. Technology has taken the music world by the throat, and there's no better tool to promote music than the Internet.
